      目的:研究β-榄香烯对DDP耐药卵巢癌细胞SKOV3/DDP增殖生长、逆转耐药的作用,以及对耐药细胞中耐药相关蛋白表达的影响。方法:CCK-8法研究β-榄香烯对SKOV3/DDP细胞增殖的抑制作用;克隆形成实验研究β-榄香烯对SKOV3/DDP细胞克隆形成的影响;流式细胞术研究β-榄香烯对SKOV3/DDP细胞凋亡的影响;CCK-8法研究β-榄香烯对SKOV3/DDP细胞产生顺铂(DDP)耐药的逆转指数;Western Blot法研究β-榄香烯对SKOV3/DDP细胞中耐药相关蛋白ABCB1、LRP和P-gp表达水平的影响。结果:β-榄香烯在20、40、80 μmol·L-1时能够抑制SKOV3/DDP细胞的增殖、克隆形成并促进细胞凋亡;β-榄香烯在不具有直接杀伤细胞作用的10 μmol·L-1时即可以逆转SKOV3/DDP细胞对DDP的耐药,其逆转耐药指数为2.58倍;β-榄香烯在20、40、80 μmol·L-1时能够减少耐药相关蛋白ABCB1、LRP和P-gp表达。结论:β-榄香烯能够直接抑制SKOV3/DDP细胞增殖、促进其凋亡并逆转细胞DDP耐药,其作用机制可能与减少耐药相关蛋白ABCB1、LRP和P-gp表达有关。

      Objective: To investigate the effect of β-elemene on the proliferation and reversal of drug resistance of SKOV3 / DDP ovarian cancer cells, and its effect on the expression of resistance associate proteins. Methods: CCK-8 method was used to test the inhibitory effect of β-elemene on the proliferation of SKOV3 / DDP cells; Clone formation experiment was used to study the effect of β-lemene on the colony formation of SKOV3 / DDP cells; Flow cytometry was used to study the effect of β-lemene on the apoptosis of SKOV3 / DDP cells; CCK-8 method was used to calculate the reversal index of DDP resistance of SKOV3 / DDP cells; Western blot was used to determine the protein expression of ABCB1, LRP and P-gp in SKOV3 / DDP cells. Results: β-Elemene inhibited the proliferation and clone formation while promoted the apoptosis of SKOV3 / DDP cells at 20, 40 and 80 μmol·L-1; β-Elemene reversed the resistance of SKOV3 / DDP cells to DDP at 10 μmol·L-1 without direct killing effects, and the reversal index was 2.58; β-Elemene reduced the expression of ABCB1, LRP and P-gp at 20, 40 and 80 μmol·L-1. Conclusion: β-Elemene can directly inhibit the proliferation, promote the apoptosis and reverse the DDP resistance of SKOV3 / DDP cells, and the mechanism may be related to the decrease of ABCB1, LRP and P-gp expression.
