      目的:建立HPLC法同时测定不同来源的红曲中腺苷、腺嘌呤、洛伐他汀的含量。方法:采用Agilent Zorbax SB C18(4.6 mm × 250 mm,5 μm)色谱柱;流动相:乙腈(A)-0.1%磷酸(B),梯度洗脱;柱温:40 ℃;检测波长:254 nm;流速:1 mL·min-1;进样量:20 μL。结果:腺苷、腺嘌呤、洛伐他汀含量的质量浓度范围分别在0.10~6.40、0.20~12.80、1.00~64.00 μg·mL-1,与各自的峰面积呈良好的线性关系(r=0.999 1、0.999 5、0.999 2)。加样回收率在95.08%~106.3%范围内,RSD均低于4.41%。结论:该方法重现性好、准确精密,可用于红曲中腺苷、腺嘌呤及洛伐他汀的定量分析。
英文摘要:
      Objective: To develop an HPLC method for the determination of adenosine, adenine and lovastatin in red yeast rice from different manufacturers. Methods: An Agilent Zorbax SB C18 (4.6 mm × 250 mm, 5 μm) column was adopted for separation at 40 ℃. Acetonitrile (A)-0.1% H3PO4 (B) was used as the mobile phase for gradient elution. The injection volume was 20 μL and the flow rate was 1 mL·min-1. The detection wavelength was 254 nm. Results: Adenosine, adenine and lovastatin were all analyzed quantitatively, the linear ranges were 0.10-6.40, 0.20-12.80 and 1.00-64.00 μg·mL-1, respectively. The values were 0.999 1, 0.999 5 and 0.999 2, respectively. The recoveries of the three analytes ranged from 95.08% to 106.3% and the relative standard deviations were all within 4.41%. Conclusions: The method is reproducible and accurate, and can be used for the determination of adenosine, adenine and lovastatin in red yeast rice.
